Genetic induction of structural changes in barley chromo- somes added to common wheat by a gametocidal chromosome derived from Aegilops cylindrica

چکیده
A gametocidal chromosome, chromosome 2C derived from Aegilops cylindrica, was introduced into six wheat-barley addition lines to establish chromosome-mutationinducing lines. These lines, except the one for chromosome 4H, had a pair of respective barley chromosomes and one 2C chromosome (22”+1’); the line for 4H was doubly monosomic for chromosome 4H and chromosome 2C (21”+2’). In these lines, chromosome breaks were expected to occur in the gametes lacking chromosome 2C. The selfed and backcrossed progeny of these lines were cytologically investigated by in situ hybridization and N-banding. Various types of structural aberration like deletion and translocation were detected for all barley chromosomes with frequencies ranging from 10.8% to 27.9%. The highest frequency of aberrations occurred in chromosome 4H with most of the breakpoints localized in or near the centromeric region. A simplified procedure of in situ hybridization used in this study was suitable for the large-scale screening for barley chromosome aberrations in the background of wheat.

Karyological studies in Triticum monococcum subsp. aegilopoides and Aegilops cylindrica species grown wild pairwise in west Iran
The wild-relative gene pools of wheat are a rich source of genetic variation for wheat improvement.Karyotypes of eight genotypes of Triticum monococcum subsp. aegilopoides and eight genotypes of Aegilops cylindrica, which grown wild next to each other in different regions in the west of Iran were studied. Root tips were treated in α-bromonaphthalene solution, fixed in chromic acid-formaldehyde f...

We used two gametocidal (Gc) chromosomes 2C and 3C(SAT) to dissect barley chromosome 4H added to common wheat. The Gc chromosome induced chromosomal structural rearrangements in the progeny of the 4H addition line of common wheat carrying the monosomic Gc chromosome. We conducted in situ hybridization to select plants carrying rearranged 4H chromosomes and characterized the rearranged chromosom...

The gametocidal (Gc) chromosome from Aegilops spp induces chromosome mutation, which is introduced into common wheat as a tool of chromosome manipulation for genetic improvement. The Gc chromosome functions similar to a restriction-modification system in bacteria, in which DNA methylation is an important regulator. We treated root tips of wheat carrying Gc chromosomes with the hypomethylation a...

Aegilops variabilis (UUSvSv), an important sources for wheat improvement, originated from chromosome doubling of a natural hybrid between Ae. umbellulata (UU) with Ae. longissima (SlSl). The Ae. variabilis karyotype was poorly characterized by fluorescent in situ hybridization (FISH). The FISH probe combination of pSc119.2, pTa71 and pTa-713 identified each of the 14 pairs of Ae. variabilis chr...

Introgression from allohexaploid wheat (Triticum aestivum L., AABBDD) to allotetraploid jointed goatgrass (Aegilops cylindrica Host, CCDD) can take place in areas where the two species grow in sympatry and hybridize. Wheat and Ae. cylindrica share the D genome, issued from the common diploid ancestor Aegilops tauschii Coss. It has been proposed that the A and B genome of bread wheat are secure ...
